Herramientas bioinformáticas in silico para identificar compuestos neuroprotectores en Alzheimer y Parkinson: una Revisión Sistemática

Abstract
Parkinson's and Alzheimer's diseases represent a growing public health challenge due to their high prevalence, irreversible progression, and the limited efficacy of current treatments. For this reason, the identification of neuroprotective compounds using in silico tools emerges as a promising strategy to optimize the discovery of new pharmacological treatments. This systematic review aims to analyze the potential of these bioinformatics tools in the development of strategies for identifying molecules with neuroprotective activity for both diseases. The methodology was developed following the PRISMA guidelines, through the search, selection, and critical analysis of scientific articles, including studies of molecular docking, molecular dynamics, and experimental validation, evaluating affinity energy and conformational stability. The results showed that for Alzheimer's disease, the main therapeutic targets were acetylcholinesterase (AChE), β-secretase (BACE1), and glycogen synthase kinase-3β (GSK-3β), with molecules such as curcumin, quercetin, and berberine standing out due to their favorable energies and interactions. In Parkinson's disease, the most studied targets were monoamine oxidase B (MAO-B) and alpha synuclein, where compounds such as resveratrol, luteolin, and baicalein showed neuroprotective potential. Based on these findings, in silico tools allowed for the identification of promising compounds and multi-target molecules; however, limited experimental validation and a lack of methodological standardization are factors that affect the reliability of the results.

Las enfermedades de Parkinson y Alzheimer representan un desafío creciente para la salud publica debido a su alta prevalencia, progresión irreversible y la limitada eficacia de los tratamientos actuales. Por esta causa, la identificación de compuestos neuroprotectores por medio de herramientas in silico surge como una estrategia prometedora para optimizar el descubrimiento de nuevos tratamientos farmacológicos. La revisión sistemática tiene como objetivo analizar el potencial de estas herramientas bioinformáticas en el desarrollo para la identificación de moléculas con actividad neuroprotectora para ambas patologías. La metodología se desarrolló bajo los lineamientos PRISMA, mediante la búsqueda, selección, y análisis crítico de artículos científicos, incluyendo estudios de acoplamiento molecular, dinámica molecular y validación experimental, evaluando energía de afinidad, estabilidad conformacional. Los resultados evidenciaron que para la patología de Alzheimer las principales dianas terapéuticas fueron la acetilcolinerasa (AChE), β-secretasa (BACE1) y glicógeno sintasa quinasa-3β (GSK-3β), destacándose moléculas como la curcumina, quercetina y berberina debido a energías e interacciones favorables. En párkinson, las dianas más estudiadas fueron la monoamino oxidasa B (MAO-B) y la alfa-sinucleína, donde compuestos como resveratrol, luteolina y baicaleína mostraron potencial neuroprotector. De acuerdo con estos hallazgos las herramientas in silico permitieron identificar compuestos promisorios y moléculas multitarget, sin embargo, la limitada validación experimental y falta de estandarización metodológica son factores que afectan la confiabilidad de los resultados
